Have you tried the suggestions at :

http://lifewiki.net/sixapart/MTPerformanceTuning

I believe there has been a great deal of work done for the latest 
versions based around improving performance.

dprofpp should also be able to outline for you generally where the 
bottlenecks are.
For example, it could be that the connection to the database is slowing 
down the response time
